چکیده

Some aspects of tricarboxylic acid-cycle activity during differentiation and aging in Dictyostelium discoideum were examined. The concentrations of glutamate, aspartate, alanine, citrate, 2-oxoglutarate, succinate, fumarate, malate, oxaloacetate, pyruvate and acetyl-CoA were determined at four stages over the course of differentiation. The rate of 02 utilization was also determined over differentiation. In addition, experiments are described in which the specific radioactivities of citrate, 2-oxoglutarate, succinate, fumarate and malate were determined during a 30min labelling of cells from the preculmination stage of development with [14C]glutamate, ["4C]aspartate or ["4C]alanine. A similar experiment was also performed with cells from the aggregation stage of development using ['4C]glutamate.

The Tricarboxylic Acid Cycle in Dictyostelium discoideum

Experimental studies of enzyme kinetics in vitro and metabolic fluxes in vivo have been used by Wright and her colleagues to develop a detailed kinetic model of the tricarboxylic acid cycle in Dictyostelium discoideum. This model has recently been been analyzed by two different methods (Albe, K. R., and Wright, B. E. (1992) J. Biol. Chem. 267, 3106-3114; Shiraishi, F., and Savageau, M. A. (199...

Dictyostelium discoideum developmentally regulated genes whose expression is dependent on MADS box transcription factor SrfA.

The MADS box transcription factor SrfA is required for spore differentiation in Dictyostelium discoideum. srfA null strains form rounded spores that do not resist adverse environmental conditions. Five genes whose expression is dependent on SrfA have been isolated by differential hybridization. One of these genes, sigC, is identical to phg1b, previously characterized in mutants with altered adh...

Dictyostelium discoideum during Growth in Axenic Culture

1. The specific activities of fl-N-acetylglucosaminidase, acid phosphatase, ac-mannosidase, P-glucosidase, UDP-glucose pyrophosphorylase and alkaline phosphatase have been determined in myxamoebae of the cellular slime mould Dictyostelium discoideum Ax-2 grown on different media and in different phases of the growth cycle. 2. Variations in enzymic composition occur with changes in growth medium...

Novel zinc protease gene isolated from Dictyostelium discoideum is structurally related to mammalian leukotriene A4 hydrolase.

The allantoicase (allC) gene of Dictyostelium discoideum allC RNAi mutant strain was silenced using the RNA interference technique. The mutant strain is motile, aggregated, and could not undergo further morphological development. The growth rate is high and the cells show a shortened cell cycle comparing with wild-type D. discoideum. However, the mechanisms regarding these actions remain unclea...

Isolation and characterization of casein kinase I from Dictyostelium discoideum.

In the present study, the molecular cloning and characterization of a 49-kDa form of casein kinase (CK)I from Dictyostelium discoideum is reported. The predicted amino acid sequence shares 70% identity with the catalytic domain of the mammalian delta and epsilon isoforms, Drosophila CKIepsilon and Schizosaccharomyces pombe Hhp1, and 63% identity with Hrr25, a 57-kDa form of yeast CK involved in...
